Genotropin (somatropin) is a recombinant human growth hormone used to treat growth failure in children and adults due to insufficient natural growth hormone production. It is indicated for pediatric patients with growth hormone deficiency (GHD), Prader-Willi syndrome, small for gestational age (SGA) without catch-up growth by age 2, Turner syndrome, and idiopathic short stature (ISS). In adults, it is used for replacement therapy in those with adult-onset or childhood-onset GHD. The medication is administered via subcutaneous injection using a specialized pen device or the Genotropin Miniquick delivery system. Dosage is individualized based on the condition, with pediatric doses ranging from 0.16 to 0.48 mg/kg/week and adult doses typically starting at 0.2 mg/day or 0.04 mg/kg/week, adjusted based on response and insulin-like growth factor I (IGF-I) levels. It is contraindicated in patients with active malignancy, acute critical illness, hypersensitivity to somatropin or its excipients, active proliferative or severe non-proliferative diabetic retinopathy, and children with closed epiphyses. Common adverse reactions include injection site reactions, lipoatrophy, headaches, and fluid retention, with monitoring required for potential complications like impaired glucose tolerance, intracranial hypertension, and hypothyroidism.
