INTERSTITIAL FLUID, CONTINUOUS GLUCOSE MONITORING, AND DIABETES

Interstitial fluid has allowed new diabetes monitoring technology to be developed.

Continuous glucose monitors (CGM) are machines that measure the glucose level within the interstitial fluid.

A small sensor is inserted just under the skin and this is left in place for a number of days.

The sensor is then able to read the glucose level within the surrounding interstitial fluid and send these readings to a display once every few minutes.

INTERSTITIAL FLUID AND BLOOD SUGAR LEVELS

The glucose level in interstitial fluid is not quite the same as that in our blood so a continuous glucose monitor will apply a formula to give a close guideline of what our blood glucose level is likely to be at that point in time.

It is important to note that glucose levels in our blood rise and fall ahead of glucose levels in our interstitial fluid.

The glucose level provided by a continuous glucose monitor will experience around a 10 minutes delay compared with the glucose level given by a blood glucose meter.

Therefore, if you are using a CGM, and you suspect your blood glucose levels are rising or falling relatively quickly, it will be better to do a blood glucose test if you need an accurate result, such as if you are experiencing the signs of hypoglycemia.

Hanks salts is a collective group of salts rich in bicarbonate ions, formulated in 1940 by the microbiologist John H. Hanks. Typically, they are used as a buffer system in cell culture media and aid in maintaining the optimum physiological pH (roughly 7.07.4) for cellular growth. Due to their poorly reactive nature and small concentration in solution, Hanks salts are mainly used in media that are exposed to atmospheric conditions as opposed to CO

2 incubation. Performing the latter drastically exceeds the buffer capacity of Hanks salts and may result in cell death.

They provide the thinnest possible support film while maintaining adequate strength and give practically no background interference in the TEM. As much of the specimen is unsupported, these films are appropriate for use where a structure free background is of importance. Lacey carbon films are particularly suitable for studying some nanotubes, acicular crystals and biological/materials specimens.

A variety of film-forming solutions are available to prepare support films and replicas of smooth surfaces.

Formvar (polyvinyl formal) is normally used as a 0.25% solution in chloroform. The thickness and stability of the film can be altered by adjusting the concentration between 0.25% and 0.5%.

Pioloform is polyvinyl butyral dissolved in chloroform (0.3 - 1.5%). The solution should be stored in a refrigerator in an amber glass bottle, and brought up to room temperature before use.

Butvar B98 is a polyvinyl butyral resin containing 20% polyvinyl alcohol, and is normally used as a 0.25% solution in chloroform. It is an alternative support film to Formvar as reported by Handley and Olsen, Ultramicroscopy4, p479 - 480, 1979. Films made from Butvar are hydrophilic, and are therefore suitable for negative staining methods, having good mechanical stability, high electron transparency and minimum intrinsic structure. Butvar B98 can also be applied to grids as an adhesive.

Formvarcoated grids without a carbon layer are suitable for TEM applications up to 100kV. The films are available on grids made of copper, nickel or gold.

Nb4AlC3 is a member of the vdW MAX layered material system. Similar to graphite and MoS2, the MAX phases are layered and have the general formula: Mn+1AXn, (MAX) where n = 1 to 3, M is an early transition metal, A is a non-metal element and X is either carbon and/or nitrogen. In this particular case, M=Nb, A=Al, and X=C with n=3.

Quartz discs, or wafers, are made from high quality fused quartz, ground and polished to optical grade on both sides. Quartz wafers have excellent chemical resistance against a wide variety of solvents, as well as exceptional heat resistance, with high dimensional stability over a wide temperature range. Cellulose acetate sheets and tape provide a quick and convenient means of replicating surfaces, particularly from large objects which cannot be sectioned. They soften in acetone, allowing them to be applied to surfaces which have been pre-wetted with acetone, then the replica can be stripped when dry. For fine detail, positive replicas can be made by coating the surface with carbon and then dissolving away the acetate sheet with acetone. The carbon replicas can then be examined in the TEM. Alternatively, the replicated surface can be covered with a conductive coating and examined by SEM as a negative replica.
